DSP-d8 deuterated crosslinking reagent, Dithiobis(succinimidyl 2,2,3,3,6,6,7,7-propionate-d8), is a cell membrane permeable, homobifunctional, protein crosslinker containing eight deuterium atoms to provide an eight Dalton shift by mass spec compared to the non-dueterated DSP (DSP-d0). DSP-d8 deuterated crosslinking thakes place via amine-reactive NHS esters at both ends of a cleavable, 8-atom (12.0 angstrom) spacer arm. The deuterated crosslinking spacer arm contains a reducible disulfide bond (SS-bond) which makes it a good choice for numerous applications where reversible crosslinking is desired.
Use the heavy DSP-d8 deuterated protein crosslinker in combination with the light, non-deuterated analog (DSP-d0) for mass spec studies.
DSP-d8 deuterated crosslinker must be dissolved in an organic solvent, such as DMSO or DMF, and then may be added to an aqueous crosslinking reaction. Since DSP-d8 crosslinking reagent is cell membrane permeable it may be used to crosslink intracellular proteins.
